You ca done this by two ways:
  1. Create custom IGridColumnHeaderRenderer implementation
  2. Use client side JS scripts

Add a "id" in a first "div" how this, "mygrid":
<div id="mygrid" style="width: 100%;">
@if (Model.Count() > 0)
    @Html.Grid(Model).Columns(columns =>
   columns.Add(c => c..........
After this create a javascript in the final of code:
<script type="text/javascript">
document.getElementById("mygrid").getElementsByTagName("div")[2] .getElementsByTagName("span")[0].setAttribute("title", "THE NEW TEXT ");
The number [2] and the number [0] in this javascript may depend on how you put the columns on your grid... This numbers represents de position of this div in the div called mygrid!

To know this, you can create a js like this and count how many divs appear, look:
<script type="text/javascript">
